ABSTRACT

Dicarabrols B and C (1 and 2), two new carabrane sesquiterpenoid dimers, along with one new carabrane sesquiterpenoid (3) were isolated from the whole plant of Carpesium abrotanoides L. Their full structures were established by extensive analysis of HR-ESI-MS and NMR spectroscopic data, and time-dependent density functional theory (TDDFT) electronic circular dichroism (ECD) calculations. ABSTRACT

Objective To study the effects of carpesium abrotanoides petroleum ether fraction on phagocytic function of macrophages and antibody forming cell in mice.Methods The extract carpesium abrotanoides petroleum ether fraction,forty five mice were divided into the control group and low and high dose groups of carpesium abrotanoides petroleum ether fraction by random number table method,15 mices in each group.The low-dose and high-dose groups of carpesium abrotanoides petroleum ether fraction were given 50 mg/kg and 100 mg/kg respectively by gavage for 10 days.The spleen weight,growth index,the phagocytosis of macrophages and the number of antibody-forming cells in peritoneal macrophages of mice were detected by chicken erythrocyte phagocytosis test and hemolytic plaque test.Results Compared with the control group,the spleen weight (192.4 ± 11.49 mg,204.6 4 10.59 mg vs.117.6 ± 10.89 mg),the growth index (6.04 ± 0.54,6.06 ± 0.40 vs.3.89 ± 0.14),antibody forming (1 216.4 ± 94.1,1 548.8 ± 86.4 vs.361.0 ± 11.7),phagocytosis percentage of macrophages (58.60% ± 2.60%,72.0% ± 3.08% vs.35.49% ± 1.64%),and Phagocytosis index (2.01 ± 0.10,2.69 ± 0.15 vs.0.37 ± 0.06) of the groups of low doses and high doses of Carpesium abrotanoides Petroleum ether fraction significantly increased (P<0.05).Conclusions The low doses and high doses of Carpesium abrotanoides Petroleum ether fraction can enhance the mice's specific and nonspecific immune function,and protect the immune system of mice.

ABSTRACT

Objective To study the terpenes from Carpesium abrotanoides and their cytotoxic activities. Methods The separations and purifications were taken by column chromatography over normal phase silica gel, preparative thin-layer chromatography and preparative HPLC. Their chemical structures were elucidated through spectroscopic analyses (NMR). The in vitro cytotoxic activities of the isolated compounds were studied by MTT method. Results Six eudesmane-type sesquiterpenes, one germacrane-type sesquiterpene, one carabrane-type sesquiterpene and one rareseco-sesquiterpenewere obtained and determined as granilin (1), telekin (2), isoalantolactone (3), 3α,5α-dihydroxyisoalantolactone (4), isotelekin (5), asperilin (6), 11(13)-dehydroivaxillin (7), carabrone (8), and Postia secoguaianolide (9), along with two monoterpenes 7,8-epoxy-9-(isobutyryloxy)thymolisobutyrate (10) and 10-hydroxy-8,9-dioxyisopropylidenethymol (11). Conclusion Compounds 9 and 10 are isolated from the genus Carpesium for the first time, compounds 3, 4, 6, and 10 are separated from C. abrotanoides for the first time. And it is the first time to report the 13C-NMR of compounds 4 and 6. Compound 1 shows cytotoxicity against HGC-27 cell and MDA-MB-231 cell with IC50 values of 18.18 μmol/L and 16.17 μmol/L. Compound 2 shows cytotoxicity against HGC-27 cell and MDA-MB-231 cell with IC50 values of 20.65 μmol/L and 17.62 μmol/L. Compound 8 shows cytotoxicity against HGC-27 cell with an IC50 value of 24.96 μmol/L.

ABSTRACT

Carpesii Fructus is the fruit of Carpesium abrotanoides L. and is recorded in the Chinese Pharmacopoeia (2015 Edition). Carpesium abrotanoides broadly distribute in China. Traditionally, Carpesii Fructus was used as a parasiticide,especially for ascariasis,pinworms and tapeworm disease. In ancient times,the Carpesium plants were used as traditional Chinese,Korean and Japanese herbal medicines for the treatment of several diseases.Carpesii Fructus was first recorded in the book"Newly Revised Canon of Materia Medica"in the Tang Dynasty of China.The original plant is Compositae Arte-misia santonica (Seriphidium cinum) from middle east Persian. At present, Carpesium abrotanoides issometimes confused with the Lappula family in species classification. In the Song Dynasty of China,"KaiYang Materia Medica"recorded that the best Carpesii Fructus was from Persian.The main compo-nents of Carpesii Fructusare terpenes,phenolic compounds,flavonoids and coumarins. Including telekin, 3-epi-isotelekin, 11β-13-dihydro-1-epi-inuviscolide, carabrone, carabrol, terpene lactone, gerilin, carpesia, valeric acid, oleic acid, linolenic acid, thirty-one alkane, sterol, etc. The chemical components isolated from whole plants of carpesia are more than 143. In clinical practice, Carpesii Fructus is mainly used as antiparasitic drugs and usually combined with other drugs since the poor efficacy as single drug. Its toxic reaction is closely related to the dose of the drug.Carpesia,asa main component of Carpesii Fructus, might lead to adverse reactions also. At present, Major issuesof Carpesii Fructusare the lack of phar-macological research,as well as lack of in-depth study on the material basis.Therefore,further studies are needed on the drug development and clinical usuage.
